- An interim update on the triplet therapy is expected at a medical conference in late 2025.
Promising Developments in Verastem Oncology’s Triplet Therapy for NSCLC
Verastem Oncology, a biopharmaceutical firm dedicated to innovating cancer treatments, announces notable preliminary clinical results from its RAMP 203 Phase 1/2 study, which explores a triplet combination therapy for patients with KRAS G12C mutant advanced non-small cell lung cancer (NSCLC). The combination therapy consists of three agents: avutometinib, sotorasib, and defactinib. As of November 21, 2024, the therapy has been administered to three patients previously treated with G12C inhibitors, and the initial findings are encouraging. Notably, two of these patients exhibit tumor reductions of at least 20% following their first scans, while all participants continue to tolerate the treatment without experiencing any dose-limiting toxicities (DLTs).
CEO Dan Paterson emphasizes that the combination of these drugs represents a significant advancement in addressing resistance mechanisms commonly seen in RAS/MAPK pathway-driven cancers. The study's design aims to evaluate the triplet's safety and efficacy rigorously before considering expansions in the ongoing doublet therapy cohorts. The trial has already completed enrollment for a treatment-naïve cohort, consisting of 28 patients treated with the doublet of avutometinib and sotorasib. Meanwhile, the cohort of previously treated patients is still actively enrolling and is projected to conclude its recruitment by early 2025.
The promising results from the triplet combination underscore Verastem's commitment to enhancing treatment options for patients suffering from KRAS G12C mutant NSCLC. An interim update is anticipated at a medical conference in the latter half of 2025, which could further illuminate the potential of this innovative therapeutic approach. With the ongoing trials, Verastem Oncology aims to make significant strides in improving patient outcomes and addressing the urgent need for effective therapies in this challenging cancer subtype.
In addition to the triplet therapy, Verastem continues to focus on expanding its pipeline of novel cancer therapies. The company is keen on completing the ongoing evaluations of the combination treatments while ensuring patient safety and optimizing therapeutic efficacy. The commitment to advancing cancer care through rigorous clinical research positions Verastem at the forefront of innovation in oncology.
