Oncolytics Biotech Expands GOBLET Trial to Enhance Treatment for Gastrointestinal Cancers

Oncolytics Biotech Inc. is making significant strides in the oncology sector with its clinical-stage immunotherapy, pelareorep, particularly through the ongoing GOBLET trial. This trial, which evaluates pelareorep for various gastrointestinal cancers, is partly funded by the Pancreatic Cancer Action Network (PanCAN). Recently, Dr. Dirk Arnold, the primary investigator, reports promising early efficacy signals that suggest pelareorep could be a transformative treatment for challenging tumors. As part of the strategy to enhance the trial's impact, Oncolytics Biotech announces an expansion of U.S. clinical sites, including Northwestern University, which is expected to facilitate faster patient enrollment and improve the robustness of the data collected.

The GOBLET trial includes multiple cohorts, with Cohort 4 focusing on patients with relapsed, unresectable squamous cell carcinoma of the anal canal (SCAC). Here, pelareorep is administered in combination with atezolizumab, achieving an overall response rate (ORR) of 33% from a sample of 12 patients. This response rate notably exceeds the 11% ORR observed with retifanlimab in similar patient populations, suggesting that pelareorep may offer a more effective treatment option. Currently, this cohort has enrolled 20 patients, with expectations to complete enrollment by late 2025, while an efficacy update is anticipated in Q4 2025.

In addition, Cohort 5 of the GOBLET trial is particularly promising, as it assesses the efficacy of pelareorep in combination with modified FOLFIRINOX, with or without atezolizumab, for first-line metastatic pancreatic ductal adenocarcinoma (mPDAC). Initial results from a previous cohort indicate a remarkable 62% ORR in 13 evaluable patients, providing a solid foundation for this ongoing research. Presently, approximately 40% of the 60-patient enrollment in this cohort is complete, with full enrollment projected by the end of 2026. An interim efficacy update is expected in Q1 2026, further positioning Oncolytics Biotech to make a significant impact in the treatment landscape for gastrointestinal cancers.

In addition to its advancements in the GOBLET trial, Oncolytics Biotech is well-positioned to leverage the regulatory momentum seen in the oncology space, especially following recent FDA approvals of new cancer treatments. The company aims to utilize clinical data not only to verify the efficacy of pelareorep but also to establish it as a platform immunotherapy for gastrointestinal tumors, where effective treatment options remain critically needed. With the global cancer immunotherapy market projected to reach $258.22 billion by 2031, Oncolytics Biotech’s innovative approaches and strategic expansions are set to play a vital role in this rapidly evolving field.
