Janux Therapeutics Advances Immunotherapy with Promising Results for JANX007 in Prostate Cancer

Janux Therapeutics, Inc., a clinical-stage biopharmaceutical company specializing in immunotherapy, reveals encouraging interim clinical data from its JANX007 program, which targets metastatic castration-resistant prostate cancer (mCRPC). As of November 15, 2024, the Phase 1a trial has enrolled patients who have undergone a median of four previous therapies. The trial evaluates the efficacy and tolerability of JANX007, with 16 patients treated at varying doses from 2 mg to 9 mg prior to receiving the PLUVICTO® treatment. The results showcase high prostate-specific antigen (PSA) response rates, with all participants experiencing at least a 50% decline in PSA levels (PSA50), and a significant portion achieving even greater reductions—63% reaching a 90% decline (PSA90) and 31% a remarkable 99% decline (PSA99).

The durability of these responses is particularly noteworthy, with 75% of patients sustaining PSA50 declines for at least 12 weeks and 50% maintaining PSA90 declines for the same period. Additionally, the trial indicates promising anti-tumor activity, as confirmed and unconfirmed partial responses are observed in 50% of patients evaluable by RECIST criteria. Importantly, the safety profile of JANX007 remains favorable; adverse events, primarily cytokine release syndrome (CRS), are mainly confined to the initial treatment cycle and are generally classified as grades 1 and 2. The maximum tolerable dose has yet to be determined, leaving room for further investigation into the drug's potential.

Encouraged by these results, Janux Therapeutics plans to progress to Phase 1b expansion trials, proposing two once-weekly dosing regimens for patients in the pre-PLUVICTO® second and third lines of therapy. President and CEO David Campbell emphasizes the potential of JANX007 to address the considerable unmet needs within this patient population grappling with advanced prostate cancer. Further updates on the program are anticipated in 2025, indicating a sustained commitment to advancing treatment options in this challenging domain.

In parallel to the trial developments, Janux Therapeutics schedules a virtual event to share these findings with stakeholders, enhancing transparency and engagement within the oncology community. As the company continues to explore the capabilities of immunotherapy in combating aggressive cancer types, the results of the JANX007 program could represent a pivotal moment in the treatment landscape for mCRPC.
