Janux Therapeutics Reports Promising Clinical Data for JANX007 in Prostate Cancer Treatment

Janux Therapeutics, Inc., a clinical-stage biopharmaceutical company specializing in immunotherapy, unveils encouraging interim clinical data from its JANX007 program, aimed at treating metastatic castration-resistant prostate cancer (mCRPC). As of November 15, 2024, the ongoing Phase 1a trial has successfully enrolled patients who have previously undergone a median of four therapies. Early results reveal that among the 16 patients treated with doses ranging from 2 mg to 9 mg, all exhibit significant declines in prostate-specific antigen (PSA) levels. Specifically, every patient achieves at least a 50% PSA reduction (PSA50), while 63% achieve a 90% decline (PSA90) and 31% experience a remarkable 99% decline (PSA99). These results underscore the potential effectiveness of JANX007 in a challenging patient population.

The trial further demonstrates notable anti-tumor activity, with 50% of patients showing confirmed and unconfirmed partial responses based on RECIST criteria. Additionally, the durability of treatment response is encouraging; 75% of the patients maintain PSA50 declines for a minimum of 12 weeks, and half of the patients sustain PSA90 declines over the same period. Importantly, the safety profile of JANX007 appears favorable, as the majority of adverse events, primarily cytokine release syndrome (CRS), are observed during the first treatment cycle and are generally classified as grades 1 and 2. Although the maximum tolerable dose has yet to be determined, the current findings provide a strong foundation for the next steps in the clinical development of JANX007.

In light of these promising results, Janux Therapeutics plans to initiate Phase 1b expansion trials, proposing two distinct once-weekly dosing regimens targeted at patients in the pre-PLUVICTO® second and third lines of therapy. David Campbell, President and CEO of Janux, expresses optimism about the potential of JANX007 to address the significant unmet medical needs in this patient demographic. The company anticipates further updates regarding the program in 2025, highlighting its commitment to advancing innovative therapies for prostate cancer patients. A virtual event scheduled for today at 4:30 PM ET will provide additional insights into these findings.

Janux Therapeutics' focus on immunotherapy positions it well in the rapidly evolving biopharmaceutical landscape, particularly in the oncology sector. With the increasing incidence of prostate cancer and the limitations of existing treatment options, the positive interim data from the JANX007 program may represent a significant advancement in therapeutic strategies for mCRPC. The company’s commitment to rigorous clinical testing and innovation reflects a broader trend in the industry towards personalized medicine and targeted therapies.
